What is it about?

The drug 5-fluorouracil is a widely used compound for treatment of various types of cancer. Its anticancer activity is increased by the addition of folinic acid, a form of vitamin B9. This combination has proven efficacy and is now currently used for treatment of various types of cancer, notably gastro-intestinal and more recently pancreatic carcinomas. We demonstrate herein that the potency of the 5-fluorouracil combined with folinic acid is augmented by the addition of Pyridoxal 5'-phosphate, a form of vitamin B6. The paper contains the biochemical rationale for this new combination and suggestion that the effect is potentially feasible in vivo.

Why is it important?

The new combination offers the potential to enhance the efficacy of 5-fluorouracil for treatment of cancer

Perspectives

Further demonstration is required in vivo, through animal tumor models and/or adequate clinical explorations in cancer patients accompanied with dose-finding and pharmacokinetic studies.
