What is it about?

Organoids are gaining widespread attention as a novel in vitro 3D model, but the 3D structure makes traditional biological characterization methods no longer applicable. In this paper, we develop a laminating device that enables organoids to overcome the problem of rapid characterization in spatial transcriptome.

Why is it important?

We used a lamination device to unfold 3D organoids into single-cell layers by controlled pressing, and then performed spatial transcriptome sequencing on a chip. We found that lamination does not completely lose positional information. We verified that the structural information of the organoid can be retained after lamination by comparing structural staining of the sections.

Perspectives

This was the first work I was responsible for, thanks to the help of my mentor and and lab friends. I hope this article will provide new methods for characterisation of organoids.
