I wanted to exploit the remarkable imine forming properties of salicylaldehydes by using it to attach a ligation auxiliary onto a peptide chain post-synthesis. We had worked on the auxiliary synthesis for a very long time and realised that we could shorten the synthesis to two easy steps. Ben was presenting his work on NEMO ubiquitylation and it seemed an ideal system to explore. He wanted to see what components of the linear ubiquitylation apparatus were used for the elongation step and he needed monoubiquitylated NEMO to define it.
