What is it about?

DNA scaffolds are critical for the assembly of DNA origami nanostructures. We purified single stranded DNA scaffolds (449 and 1000 nt long) using magnetic beads functionalized with short (20 nt) oligonucleotides. The purification technique is superior to the standard extraction from agarose gels. Further, the beads can be reused.

Why is it important?

Scaffolds are usually purified by agarose gel extraction, a laborious, time consuming, limited, and nonscalable technique that presents low recovery yields, delivers low-quality products, and requires specific equipment. The scalable affinity-based method presented here, which comprises magnetic particles and a simple magnet, facilitates DNA scaffold purification, thus contributing to the development of DNA origami applications.
