What is it about?

RNAi has emerged itself as a huge potential therapeutic approach to target different genetic and rare diseases and various types of cancers. Different physiological barriers are a major hurdle for delivering functional RNA materials into the cytoplasm. This review discusses about the different examples and the designing principles used in constructing different classes of delivery vehicles which can overcome these physiological barriers and result in silencing of a desired gene, with a special emphasis on peptide based delivery vehicles. This review also discusses the role of RNAi in different epigenetic modifications, immunomodulation and combination modalities.

Why is it important?

This study discusses the hallmark characters and designing principles which can be explored to construct a successful delivery vehicle for RNAi therapeutics. Examples of RNAi in epigenetic modifications, immunomodulation and combination modalities can be helpful in developing a patient specific personalized therapy against cancer. Lastly, this review present diverse opportunities and also discusses about different strategies which can be employed in future to develop efficient delivery vehicles with a potential to be translated into clinics and can also prove to be helpful in developing improved mRNA vaccines against COVID-19 .
